Sensitive Quantitation of Double-Stranded DNA in Solution

Simple, Accurate DNA Quantitation. No Dilutions. No Extra Tubes.

The QuantiFluor® ONE dsDNA System provides a fluorescent double-stranded DNA-binding dye (504nm Ex/531nm Em) in an automation-friendly, “Add-and-Read” format for both dye and standard, simplifying DNA quantitation and speeding up your workflow. It’s as easy to use as NanoDrop® absorbance-based methods, but much more sensitive for low-concentration samples.

Improved Sensitivity vs. Absorbance and Other dsDNA Fluorescent Quantitation Systems

The QuantiFluor® dsDNA dye offers greater sensitivity over absorbance at 260nm (NanoDrop) low-concentration samples, and better or equal sensitivity performance when compared to Quant-IT™ dsDNA BR.  QuantiFluor® ONE dye sensitivity range is 0.2–400ng/µl (1µl sample in 200µl assay).

Sample* Assay
Quantifluor® ONE dsDNA System 0.2–400ng/µl 0.2–400ng
Qubit® dsDNA HS 0.2–100ng/µl 0.2–100ng
Qubit® dsDNA BR 2–1,000ng/µl 2–1,000ng

*Based on 1µl sample input per assay.

DNA Quantitation Assay Sensitivity and Linearity

The QuantiFluor® ONE System detects as little as 1ng/ml dsDNA per assay using the Quantus™ Fluorometer (corresponding to 1µl of a 200ng/ml starting sample). Detection limit is defined as greater than three standard deviations above background RFU.

These graphs show representative QuantiFluor® ONE dsDNA System standard curves generated from Lambda or K562 DNA standards in a 96-well plate format. The assay linearity and dynamic range of either the QuantiFluor® ONE Lambda DNA or Genomic K562 DNA standards are shown from a range of 0.2–400ng/µl (1µl sample in a 200µl assay).

How do fluorescent dyes compare to other quantitation methods?

In these short videos, Promega Applications Scientist Doug Wiezorek explains how fluorescent dye-based methods compare to other popular quantification techniques. Check them out for some quick guidelines on choosing the right nucleic acid quantitation method for your needs.
