ReliaPrep™ FFPE gDNA Miniprep System provides a convenient method for purifying genomic DNA from formalin-fixed paraffin-embedded (FFPE) tissues. Extraction of DNA from FFPE tissues is challenging because the formalin fixation process results in cross-linking between proteins and DNA, as well as between different strands of DNA. This nonoptimal preservation of genomic DNA in FFPE complicates its use in many standard downstream analysis applications.
The ReliaPrep™ FFPE gDNA Miniprep chemistry is optimized to partially reverse this cross-linking without the need for an overnight digestion. The protocol also incorporates a deparaffinization method that does not use xylene or other hazardous or volatile solvents.
The protocol is complete in 2.5 hours with minimal hands-on time.
Purified FFPE DNA is Compatible with Sensitive Assays
Here, genomic DNA was purified from 10µm human liver FFPE sections using the ReliaPrep™ FFPE gDNA Miniprep System, and STR analysis performed using the Microsatellite Instability (MSI) Analysis System v. 1.2 (Cat.# MD1641).
